Evaluating the Status of the Sacramento Mountain Prickly Poppy.

Argemone pleiacantha ssp. pinnatisecta is a federally listed endangered subspecies endemic to a few canyons on the western slope of the Sacramento Mountains in Southern New Mexico. Three close relatives, A. squarrosa, A. polyanthemos, and A. pleiacantha ssp. pleiacantha, occur in the same broadly defined area of NM, and these are all morphologically distinguished by relatively small differences in morphology. The nature of these differences and the lack of a recent comprehensive treatment for the group have led to questions regarding the taxonomic status of A. pleiacantha ssp. pinnatisecta and consequently its endangered status designation. The aim of this research is to address the taxonomic uncertainty surrounding the taxon by applying highly variable AFLP markers to A. pleiacantha ssp. pinnatisecta and its close relatives. A total of 94 individuals representing the A. pleiacantha ssp. pinnatisecta and geographically proximate representatives of each of the aforementioned relatives were sampled. Preliminary analyses using selective primer combination EcoRI-AC and MseI-CTA and PCO identify that A. pleiacantha ssp. pinnatisecta is more distinct from all sampled congeners than the others are from each other. In addition, preliminary results suggest that A. pleiacantha ssp. pinnatisecta shows fixed allelic differentiation relative to all other samples, which is consistent with A. pleiacantha ssp. pinnatisecta being reproductively isolated, supporting its recognition as a distinct taxon.
