Pattern of genetic diversity and introgression among two endemic eusporangiate ferns, Archangiopteris somai and itoi, and wide distributed eusporangiate fern, Angiopteris lygodiifolia in.

Archangiopteris somai Hayata and Archangiopteris itoi Shieh are rare and endemic ferns in Taiwan. Only two populations of each species are restricted in Wulai and Lienhwachi, in northern and central Taiwan respectively. Angiopteris lygodiifolia is a common fern in ravines distributed in Japan, the Ryukyus and Taiwan. In this study, we estimate the genetic variation and differentiation among species, within and between populations of these fern using sequences of the rbcL-atpB noncoding spacer of the cpDNA and the internal transcribed spacer (ITS) in the nuclear ribosomal DNA. In the cytological evidence, Archangiopteris somai is a tetraploid, Archangiopteris itoi is a triploid and Angiopteris lygodiifolia is a diploid species. Using the result of the maternal DNA, we get evidence to show the Archangiopteris itoi shared haplotypes either belong Archangiopteris somai or Angiopteris lygodiifolia. This evidence suggested the Archangiopteris itoi might be a bidirectional hybridized species. In the result of the nuclear DNA, we get two types of ITS in the nuclear ribosomal DNA. The type I shared among three species and type II is a specific group in Archangiopteris genus. Combined cpDNAand nrDNA result, it was suggested Angiopteris lygodiifolia is the ancestral species of Archangiopteris somai and Archangiopteris itoi. Archangiopteris itoi is a hybrid between Angiopteris lygodiifolia and Archangiopteris somai. However, Archangiopteris somai is a tetraploid and one lineage comes from Angiopteris lygodiifolia and one lineage might be extinguished.
